Introduction of Cholera

Cholera is a severe acute diarrheal illness in humans caused by different types of Vibrio cholerae. Cholera is a toxin-mediated disease in which V. cholerae expresses cholera toxin (CT) on the intestinal surface causing symptoms of profuse secretory diarrhea. However, studies have shown that immune responses to CT do not confer effective protection against cholera. Cholera is mainly spread through food or water contaminated with cholera bacteria, undercooked shellfish is also a common source of illness.

Cholera affects millions of people worldwide, especially in Africa and Southeast Asia. Currently, humans are the only known host of V. cholerae. Till now, oral rehydration salts (ORS) remain the leading treatment for cholera. An oral cholera vaccine approved by the US Food and Drug Administration (FDA) provides reasonable protection for about six months.

Symptoms of Cholera

The symptoms can range from none to mild to severe to life-threatening.

  • Large amounts of watery diarrhea
  • Vomiting
  • Muscle cramps
  • Severe diarrhea
  • wrinkling of the hands and feet
  • Sunken eyes and cold skin

Anti-Cholera Antibody

A series of antibodies against O-specific polysaccharide (OSP) of V. cholerae have been developed. These OSP-specific antibodies may prevent V. cholerae in the intestinal lumen through mechanisms such as direct bactericidal, binding, or agglutinating activities. In addition, bivalent conjugation of OSP-specific antibodies confers immune protection against cholera through motility inhibition of V. cholerae. In one study, polyclonal antibodies from the plasma of recovered cholera patients produced an antibody response to V. cholerae O1 OSP. These antibodies were able to significantly reduce the motility of the V. cholerae organism, as observed under a microscope.
