Introduction of Anti-Bacteria Abs

Ubiquitous in nature, bacteria constitute a large domain of prokaryotic microorganisms. Bacteria also have symbiotic and parasitic relationships with plants and animals and play important roles in many stages of the nutrient cycle. However, some types of bacteria are pathogenic and cause infectious diseases.

The discovery of antibiotics and the development of vaccines have greatly extended the human lifespan and improved the quality of life, but the emergence of antibiotic-resistant superbugs has brought new problems and potential challenges. Furthermore, the development of new antibiotics is lagging behind the emergence and spread of antibiotic resistance. In this case, monoclonal antibodies (mAbs) as novel therapeutic agents have been used to ameliorate infection with remarkable clinical success in several fields. Compared with polyclonal antisera, mAbs always present higher specificity, lower variability, and less cross-reaction.

Mechanisms of Anti-Bacteria Abs

In the treatment of bacterial infections, monoclonal antibodies can act through a variety of different mechanisms, including neutralization of toxins, inhibition of virulence factors, complement-mediated killing activity, and opsonophagocytosis. Till now, three mAbs have been approved by FDA for the treatment of bacterial infection.

  • Toxin neutralization - Anti-toxin antibodies block the binding of the toxin to the receptor thereby preventing structural changes of the toxin and facilitating the formation of immune complexes.
  • Inhibition of virulence factors - Antimicrobial monoclonal antibodies can target virulence factors on the surface of bacteria to hinder pathogenicity.
  • Antibody-mediated complement-dependent killing activity - The Fc region of the antibody can bind to C1q, triggering the classical complement pathway to lyse bacteria.
