Introduction of Plague
Plague is a severe infectious disease caused by the bacterium Yersinia pestis. Depending on the site of impact and associated symptoms, plague can be divided into three forms, pneumonic, bubonic, and septicemic plague. Plague has killed more than 200 million people in human history. Currently, antibiotics and supportive care remain the mainstay of treatment for plague infection. Studies have shown that scientific treatment can reduce the risk of death by 10%. However, a series of multi-antibiotic resistant strains of Yersinia have been discovered and isolated. Capsid F1 protein and the low-calcium response (Lcr) V-protein are two major virulent factors of Y. pestis that have served as important targets for immune response.
Diagnostic of Plague
Because the symptoms of plague are nonspecific, laboratory testing is required for a definitive diagnosis of plague. Identification of Yersinia by microscopy and culture samples, polymerase chain reaction (PCR)-based genetic testing of the bacteria, and blood tests for Y. pestis antibodies can be used in the diagnosis of plague.

Anti-Plague Antibody at Creative Biolabs
It has been reported that mouse anti-plague monoclonal antibodies (mAbs) can protect mice from the plague. Therefore, mAb-based therapy may replace antibiotics as an emerging therapeutic approach. Humanization of anti-plague mAbs and the development of fully human anti-plague mAbs are critical to avoid potential immunogenicity. In one study, three mAbs were isolated from a large native human phage-displayed Fab library. The combined use of these antibodies showed a clear synergistic effect, which means a major breakthrough for antibodies as adjuvant therapy for human Y. pestis infection.
