Antibody Production in Microbial Hosts

Monoclonal antibodies (mAbs) and antibody fragments present important roles in basic biological research as well as in the diagnosis and treatment of diseases. Mammalian cells are most commonly used to produce mAbs due to their ability to undergo post-translational modifications (PTMs), but some shortcomings in bioprocessing still need to be overcome. In this case, the use of microorganisms to produce antibody fragments becomes a good alternative. Here, we summarize three main microbes, including S. cerevisiae, P. pastoris, and E. coli.

  • S. cerevisiae

S. cerevisiae was first used in the production of recombinant proteins, and several drugs produced by this yeast have been successfully marketed. In particular, the production of single domain antibody (sdAb) using S. cerevisiae ensures production titers as high as hundreds of mg/L. There are three main types of shuttle vectors used for S. cerevisiae transformation, such as east episomal plasmids (Yep), yeast centromeric plasmids (Ycp), as well as yeast integrative plasmids (Yip). In recent years, a series of methods have been developed to further increase the secretory capacity and productivity of S. cerevisiae.

  • P. pastoris

P. pastoris is an alternative to S. cerevisiae for the production of mAbs and antibody fragments. Compared with S. cerevisiae, P. pastoris prefers respiratory than fermentative growth to obtain high cell densities using the cheap medium. It is important to note that extensive non-homologous recombination is a major defect of P. pastoris. Currently, two recombinant therapeutic antibody fragments using P. pastoris as the production host system are already on the market.

  • E. coli

E. coli is the first microorganism used to produce recombinant biopharmaceuticals, and currently nearly 40% of marketed drugs are produced by E. coli. Its advantages as an expression system for recombinant proteins lie in its low production cost, ability to reach high cell densities, simple genetics, and ease of operation. In general, protein expression using E. coli is performed in stirred tank reactors (STRs).
