BBB Spheroid Models

To date, human immortalized cell-based multicellular spheroidal BBB models (hiMCS-BBB models) are widely used in BBB research. Human brain endothelial cells (ECs), pericytes, and astrocytes are essential for the BBB structure. The hiMCS-BBB consisting of human immortalized brain ECs, astrocytes, and brain pericytes are co-cultured under low-adherence conditions. Among the main types of BBB models, like transwell 2D models, hydrogel models, hollow fiber models, and microfluidic models, BBB spheroids stand out with their distinguished advantage of closely mimicking the real blood-brain barrier. This showcases their significance in providing advanced and innovative insights into the blood-brain barrier function.

Advantages

The spheroid model offers several advantages over other in vitro models, including:

  • More physiologically relevant environment.
  • Larger surface area for cell-cell interactions.
  • Easier manipulation, and a more accurate representation of complex cell-cell interactions and structures.
  • An average diameter ranging from 200 to 400 μm, makes them suitable for high-throughput applications since they can be analyzed for fluorescence studies as a whole mount without the need for prior cutting.

Applications of BBB Spheroid Models in Pre-Clinical Study

In vitro BBB models enable transport and visualization studies, drug transporter functionality analysis, drug interaction and targeting studies, pharmacological safety assurance, examination of BBB functions relevant to disease, and drug discovery research.
