Construction of 3D Normal Organoid Models

In recent decades, as organoid technology develops rapidly, 3D normal organoid models have been constructed from embryonic stem cells (ESCs), induced pluripotent stem cells (iPSCs), and adult stem cells by utilizing different cell isolation operations and growth factor combinations. The donor tissues are healthy and mainly include the pancreas, stomach, colon, small intestine, and so on.

Advantages of 3D Normal Organoid Models

As an emerging ex vivo model, 3D normal organoids offer the tool ensuring the implementation of aiming at tumor cells without damaging healthy cells. Also, 3D organoids have displayed incomparable advantages over the previous modeling approaches.

  • Firstly, it’s feasible and convenient to perform CRISPR-based gene engineering on these models by introducing any combination of cancer gene alterations.
  • Secondly, there are many kinds of cells in the organoid system, which retain the in vivo microenvironment to some extent.

  • Finally, they can preserve genetic stability, and the model establishment cycle is highly efficient.

Applications of 3D Normal Organoid Models

Since its birth, the 3D normal organoid model has been seen as a powerful supplement to existing preclinical models. Generated from healthy donor tissues, they are suitable for drug toxicity assessment including hepatotoxicity, cardiotoxicity and nephrotoxicity testing. In addition, they can be applied in cancer modeling and provide new insights into the study of tumor progression and screening personalized drugs for precision medicine. More importantly, they are promising for regenerative medicine. For instance, healthy organoids generated by modern genome-editing technology in an in vitro setting can be subsequently transferred to patients for organ transplantation.
