Human γδ T cells play unique and critical roles in stress monitoring, immunity to infection, and carcinogenesis. γδ T cells have a unique advantage used in cell therapy as they can be independent of human leukocyte antigen (HLA) restriction to identify and kill transformed cells. CAR-γδ T cells that express CAR molecules that target surface antigens on tumor cells to mediate a series of tumor elimination, which will be a huge innovation in tumor therapy.

Characterization of γδ T cells
γδ T cells are promising host cells for CAR constructs with many features:
CAR-γδ T cells for Cancer Immunotherapy
CAR-γδ T cells usually present two-fold antitumor activity after transfection which is mainly by two facets, such as not only via the endogenous γδ TCRs, but also via the CAR specificity. In general, CAR-transduced Vδ2T cells have an increased cytotoxicity efficacy target to relevant tumor cells. Activated Vδ2T cells can also act as APCs and cross-present tumor-derived antigen peptides to CD8 αβ T cells while killing tumor cells. More importantly, CAR-transduced Vδ2T cells retained the ability to cross-present tumor antigens to αβ T cells.
