Normal Kidney Organoid Model

Due to its increasing prevalence and incidence across the globe, chronic kidney disease (CKD) is gradually becoming a global health problem. Unaffordable treatment hinders the timely control of many patients' conditions and leads to millions of death. If the disease progresses to end-stage renal disease (ESRD), there will be two limited treatment options including hemodialysis and kidney transplantation. Therefore, more effective and economically feasible treatment strategies need to be explored. Construction of Normal Kidney Organoid Models

For the generation process of normal kidney organoid systems, Grassi et al. have proposed a practicable protocol, as shown in Fig.1. This is the first try of culturing the organoids from both healthy kidney and clear cell renal cancer (ccRCC) tissues starting from undifferentiated enriched heterogeneous patient cultures. Their data indicates a broad range of possibilities that normal kidney organoid models can be applied in basic and translational research.

Advantages of Normal Kidney Organoid Models

Compared with traditional models, normal kidney organoids have their own unique characteristics and show exciting advantages in many aspects.

  • Display obvious similarities to normal organ development, such as cellular complexity, transcriptional congruence, and tissue morphology reproducibility.
  • Have paralleled cell types to the native organ, which has been demonstrated by single-cell RNA sequencing technology.
  • Can be used as an alternative protocol to generate induced pluripotent stem cells (iPSC)-derived organoid models with gene mutations, through the application of gene editing tool.
  • Preserve genome and phenotype stability in the long-term in vitro maintenance process.
  • Be sensitive to answer external stress and injury by expressing the corresponding biomarkers, which are very evaluable for toxicity testing and disease modeling.

Applications of Normal Kidney Organoid Models

Organoids are useful tools for establishing glomerular and tubular disease models. Based on their great potential in drug screening, enhancing the understanding of kidney disease, and renal tissue transplantation, normal kidney organoids are expected to accomplish greater breakthroughs in CKD research in the future. Some novel technique tools including gene editing, microfluidic technology and the generation of interspecies chimeras will accelerate the maturation of the organoid culture system, further expanding the application area.
