Construction of Normal Stomach Organoid Models

Normal stomach organoids are cultured from healthy cells in patient-derived tumor samples. Common construction methods are seeding gastric glands from human resection samples in a basement matrix and culturing them in a special medium containing various growth factors and inhibitors into organoids. In addition, these organoid models can also be constructed through co-culturing operations, for example, the air-liquid interface (ALI) technique.

Advantages of Normal Stomach Organoid Models

Through the current reports, normal stomach organoid models show marked advantages over current models.

  • Preserve growth behavior and morphological phenotype in the ex vivo culture process for up to a year.
  • The growth characteristics are not easily influenced or changed by freeze and thaw cycles seemingly.
  • The phenotype of normal organoids is reversible, specifically, it can be induced into malignant or normal states, which makes them suitable for studying tumorigenesis.
  • Can be employed for drug sensitivity testing and need a shorter time and a cheaper cost than animal models.

Applications of Normal Stomach Organoid Models

In stomach cancer research, normal organoid models offer a novel tool to study phenotypic consequences resulting in tumor development. Besides, they make it possible to quickly test drugs before conducting animal experiments. What needs to be emphasized is that extensive operations can be implemented in these models because they bear non-mutated genomes and potential of expressing a full spectrum of authentic phenotypes. So, a variety of stomach research will benefit from the organoid models, such as infection biology, mucosal immunology, as well as cancer research, and so on.
