What is a Current Clamp?

A current clamp, an electrical device with jaws, is a common technique in electrophysiology. It measures the current in an electrical conductor by clamping it without touching the wire directly or disconnecting it in order to insert a probe. Current clamp recording is a type of intracellular recording that entails continuously injecting positive or negative current (as "square" d.c. pulses) into the cell while measuring the voltage difference across the cell's membrane.

Applications of Current Clamp Assay

Applications Read alternating current (AC) magnitude and be able to monitor waveform and phase with supplementary devices. Some clamp meters can measure currents as high as 1000 A.
Control the current injected through the patch pipette while simultaneously recording the cell's free-running membrane potential, in addition to measuring action potentials that occur spontaneously or in response to injected stimulating current.
Measure the action potentials, postsynaptic potentials, as well as after potentials of the neurons of interest to determine how a particular neuron is stimulated or analyze the behavior of membrane potential.
Study how a cell reacts when an electric current flows through it, this is crucial for example for understanding how neurons react to neurotransmitters that function by opening membrane ion channels.

Advantages of Recording in Current Clamp

  1. The recording method has no impact on the membrane potential, and the experiment requires fewer changes as a result.
  2. The amplifier records the voltage produced by the cell on its own or in response to stimulation, and the membrane potential is free to change.
  3. This recording of a neuron firing as a result of being depolarized by current injection is made using a whole-cell current clamp.
