PDX-Derived Colorectal Cancer Organoid Model

Colorectal cancer (CRC) is a digestive malignant tumor with a dismal prognosis and low survival rate. Nowadays, few therapies are radical for the advanced CRC patient population. Therefore, requirements continue to rise for new therapeutic strategies. At Creative Biolabs, we have built a professional team to study new 3D modeling technologies such as PDX-derived colorectal cancer organoids to serve our new and existing customers.

Shortcomings of Common Models

For a long time, cancer cell lines and tumor mouse models are commonly used to solve the clinical problem of CRC research. They represent the partial characteristic but fail to capture the key properties of primary tumors including invasion and metastasis, which restricts their further development and applications. A new model that can complement each other to current models is expected by the academic world.

Advantages of PDX-derived Colorectal Cancer Organoid Models

PDX-derived tumor organoids emerge in this CRC research tide and have gradually been integrated into basic research and clinical research, influencing science horizons. Overall, they possess unique advantages over common models in terms of the following aspects.

  • Expand rapidly in a relatively short time while still keep their genetic stability.
  • Retain tumor microenvironment (TME), heterogeneity as well as patient diversity.
  • Huge high-throughput scalability for drug combination screenings.
  • Easy to perform genetic modifications in such models, for instance, CRISPR-mediated editing.
  • Offer a possibility for co-culture assays with autologous stromal components and immune cells.

Applications

An increasing collection of evidence has demonstrated that PDX-derived organoids are excellent tools worth further developing and optimizing. In the CRC research area, they show a bright prospect to boost tumor drug discovery and reduce the deviation between basic research and clinical research. Scientists combine the PDX-derived CRC organoid models with other biology methods to deal with various academic issues.
