Overview of PDXO Tumor Models

The PDXO tumor model falls into an in vitro model that is constructed from the tumor tissues of the patient-derived xenograft (PDX) models. Usually, tumor tissues are firstly dissociated into small fragments. Then, they need to be washed and filtered to remove the debris. Next, we collect and suspend the cell clusters in the basement membrane extract (BME) solution. Finally, the mixture is transferred into a culture dish and cultured in a CO2 incubator to generate organoids for subsequent drug discovery applications.

Advantages of PDXO Tumor Models

PDXOs offer significant advantages over standard 3D models and in vivo PDX models.

  • PDXO cultures keep heterogeneity and phenotypes after undergoing long-term cultivation.
  • PDXOs have the ability to preserve the genomic characteristics of original tumors and display aggressive growth dynamics.
  • Such models highly reproduce molecular and histopathological characteristics of the tumor in vivo.
  • PDXO models are amenable to high-through drug screening and offer a relatively accurate prediction of drug possible responses in vivo.
