PDX-Derived Mesothelioma Organoid Model

For malignant mesothelioma, there is no effective radical treatment strategy nowadays. Most patients are diagnosed at an advanced stage, so palliative surgery is the only helpless choice. How to overcome the current treatment challenge is worth exploring in depth. Shortcomings of Common Models

Common 2D cell cultures are planar systems, which can’t reproduce the 3D in vivo microenvironment of tissues. Due to their unfaithful reflections of drug diffusion kinetics and cell-cell/cell-matrix interactions, the drug doses often encounter failure when transformed from 2D cell culture to clinical patients. Animal models such as patient-derived xenografts (PDXs) have opened the feasibility window for studying the progression mechanism of tumors in vitro. However, they must be established on immunodeficient mice, which disturbs natural cell states and become the most fundamental limitation.

Advantages of PDX-derived Mesothelioma Organoid Models

Due to low diagnostic accuracy and shortage of a reliable model, current research in the mesothelioma field is not very plentiful. As a new alternative model for the investigateion of mesothelioma, PDX-derived mesothelioma organoid model has a few advantages, which including but not limited to:

  • They will permit mimicking tissue microenvironments, including 3D architecture, cell-cell/cell-matrix interactions and in vivo circulation.
  • As a novel in vitro 3D model, they will be able to reproduce the function of in vivo tissues.
  • They will adapt to different culture conditions and retain histological and mutational profiles of primary tumors.
  • They will obtain the ability to represent the tumor phenotypes to a certain degree.

Potential Applications

Although published data that support applications of PDXO in the investigation of mesothelioma is rare, we have evaluated the potential of PDX-derived mesothelioma organoid model in multiple fields. We believe PDX-derived mesothelioma organoid model will be extensively applied in high throughput screens, drug testing, disease modeling and precise treatment.
