Cluster of Differentiation 80 (CD80)
Cluster of differentiation 80 (CD80), also called B7, B7-1 or BB1, with a molecular weight of 60 kD, is expressed on activated B lymphocytes, activated T lymphocytes, macrophages, peripheral blood mononuclear cells, and dendritic cells. It belongs to the immunoglobulins superfamily (IgSF), and its receptors are CD28 and CD152 (CTLA4). CD80 is an essential membrane antigen for the activation of T lymphocytes and a co-stimulatory factor for CD86-activated T lymphocytes. It plays an important role in autoimmune monitoring, humoral immune response, and transplantation response. Studies have implicated that the expression levels and the ratio of CD80 and CD86 can reflect the activation status of the macrophage. Besides, CD80 exerts a potential role in the T helper cell differentiation.

The Role of CD80 in Sepsis
Sepsis, defined as the systemic inflammatory response to infection, is a life-threatening condition. It arises when the body's response to infection causes injury to its own tissues and organs. The host inflammatory response in sepsis is complex, involving an interplay between pro- and anti-inflammatory mediators, both of which have divergent effects on pathogen control, systemic inflammation, and susceptibility to recurrent infection.
A large body of evidence points to a critical role for the CD28-CD80/86 system in regulating inflammation, autoimmune disease and graft vs. host disease in the adaptive immune response. The ability of CD80 to regulate inflammation can be in part explained by its known ability to regulate the induction of NF-κB in response to CD28 engagement. Specifically, the expression CD28 in neutrophils activates macrophages in a contact-dependent manner via engagement of CD80/86. In turn, CD80/86 signal via NF-κB induces numerous cytokines, most notable IL-6. It is reported that sepsis was associated with increased levels of CD80. Anna Nolan’s study revealed that CD80 was upregulated in circulating monocytes in septic humans. And the higher CD80 levels associated with both the presence of shock and correlating with overall severity of illness as determined by SOFA score. It is supposed that future therapies targeting in sepsis could be directed specifically at CD80.
