Introduction of HLA-G5 Protein
HLA-G histocompatibility antigen, also known as human leukocyte antigen G (HLA-G), is a protein that encoded by the human HLA-G gene. As a member of the nonclassical human leukocyte antigen (HLA) family, HLA-G is structurally diverse with many different isoforms, including HLA-G1~7. Indeed, the HLA-G5, -G6, and -G7 isoforms are highly unusual, as they are spliced variants of the HLA-G mRNA retaining introns 4 and 2. Initially, the expression of HLA-G was restricted to the maternal-fetal interface, on extravillous cytotrophoblasts. While in nonfetal healthy subjects, the HLA-G protein is found only on the cornea, thymic medulla, nail matrix, beta cells of the islets of Langerhans, mesenchymal stem cells, and endothelial precursors.

HLA-G5 Protein Marker of Sepsis
Sepsis is a leading cause of death in intensive care units across the world. Studies have suggested that HLA-G is able to modulate both adaptive and innate immunity by interacting with T or B lymphocytes and NK cells or polymorphonuclear cells. And HLA-G can inhibit all types of immune competent cells by the direct binding of both completely soluble and membrane-bound isoforms to inhibitory receptors via the α3 domain. Most importantly, the plasma level of HLA-G5 protein in patients with sepsis is significantly higher than that of normal people. Moreover, persistent HLA-G5 expression in septic shock is associated with survival. Under this condition, plasma level of HLA-G5 can be used as a potential marker for the detection of sepsis.
