Signaling Pathways of Complement C5a

The complement-activated product, complement C5a, is a glycoprotein with a 74 amino acid. Upon activation of the complement cascade, C5a acts as a potent pro-inflammatory mediator cleaved enzymatically from its precursor. The C5a receptors allow C5a to elicit a broad range of effects, which are widely present throughout the body. Thus, C5a has been regarded as a significant pathogenic driver in a number of immuno-inflammatory diseases and makes C5a inhibition an attractive therapeutic strategy. C5a/C5aR signaling pathway can cooperate with TLR-4 activation by LPS on macrophages. The complement system not only represents an effective innate immune mechanism of host defense to eradicating microbial pathogens, but it is also widely involved in many forms of acute and chronic inflammatory diseases including sepsis, acute lung injury, ischemia-reperfusion injury, and asthma, to give just a few examples.

Complement C5a Marker of Sepsis

C5a often functions as a powerful inflammatory mediator and play an important role in the pathological development of many inflammatory diseases including the complement system such as sepsis, rheumatoid arthritis, inflammatory bowel disease, systemic lupus erythematosus, psoriasis. As a strong chemoattractant, C5a is involved in the recruitment of inflammatory cells such as neutrophils, eosinophils, monocytes, and T lymphocytes, inactivation of phagocytic cells and release of granule-based enzymes and generation of oxidants, all of which may contribute to innate immune functions or tissue damage. In summary, C5a has been known as a vital bridge between innate and adaptive immune functions, extending the roles of C5a in inflammation. Related research has shown that the cellular and molecular mechanisms of C5a in the development of inflammatory disorders, sepsis, acute lung injury, ischemia-reperfusion injury, and asthma.
