Macrophage Inflammatory Protein 1
Macrophage Inflammatory Proteins (MIP) belong to the family of chemotactic cytokines referred to chemokines. MIP has two major forms in humans MIP-1α and MIP-1β, also termed CCL3 and CCL4 respectively. While stimulated with bacterial endotoxin, both of them are major factors produced by macrophages. There are two important findings recently that led to a renewed interest in MIP-1 proteins, highly related ligands and activators of CC chemokine receptors (mainly CCR1, 3, and 5): (1) the effector chemokines CCL3 and CCL4, which are produced and secreted by activated macrophages to attract other pro-inflammatory cells, are also crucial in recruiting themselves to sites of inflammation; and (2) CCL3 can potently inhibit the macrophages uptake of HIV-1 via CCR5 ligation.

The Role of MIP-1 in Sepsis
It is reported that MIP-1 proteins are key players in the pathogenesis of many inflammatory conditions and diseases including sepsis, asthma, granuloma formation, wound healing, arthritis, multiple sclerosis (murine EAE model), pneumonia, and psoriasis. In septic patients, levels of MIP-1alpha and MIP-1beta were detected elevated within 24 h of sepsis. MIP-1alpha and MIP-1beta increase during acute inflammation but are not associated with fever in endotoxemic humans during cyclooxygenase blockade. MIP-1 plays a key role in the induction and modulation of inflammatory responses, i.e. in the context of autoimmune reactions and host defense responses. In addition, MIP-1 proteins regulate various aspects of tissue homeostasis. MIP-1 also has important physiological functions such as in thymic and stem cell development, angiogenesis, and tumor biology, which are discussed in excellent recent reviews.
