General Description of Elastase
Elastase (HLE) is a serine protease digest and degrades a number of proteins including elastin which is an elastic substance in the lungs and some other organs that supports their structural framework. HLE is produced in the pancreas as an inactive zymogen and activated in the duodenum by trypsin. The following information applies to porcine elastase. Elastases are defined by their ability to release soluble peptides from insoluble elastin fibers by a proteolytic process. Elastase is another protease that induces disintegration of the axoneme by microtubule sliding in the presence of ATP. Elastase and trypsin differ in their effects on the flagellar waveform of reactivated sperm, suggesting that they disrupt different structural components. It has been shown that elastase disrupts tight junctions, cause proteolytic damage to tissue and break down cytokines.

Elastase and Sepsis Disease
Sepsis is a systemic inflammatory response to known or suspected infection and associated with microvascular dysfunction and capillary leak. The cause of capillary leak may be due to substances released by activated polymorphonuclear neutrophils (PMNs) that adhere to microvascular endothelial cells. Following binding, PMN often degranulates releasing azurophilic enzymes like HLE and proteinase 3 (PR3)) at the endothelial surface and into plasma. These enzymes, particularly HLE and PR3, contribute to the loss of endothelial integrity. Through a mechanism involves the actions of HLE and PR3, neutrophil degranulation such as increased plasma concentrations can result in increased endothelial permeability. Neutrophil elastase plays an important role in the sepsis-induced myocardial dysfunction.
