Ferritin
Ferritin is a hollow globular protein with the molecular weight of 474 kDa. It consists of 24 subunits forming a nanocage with multiple metal-protein interactions. Ferritin is a highly conserved protein between species and plays an important role in many biological processes. Ferritin has been revealed to be responsible for storing iron in a non-toxic form, depositing it in a safe form, and transporting it to areas where it is required. Besides, vertebrate ferritin also possesses ferroxidase activity that can converse iron from the ferrous (Fe2+) to ferric (Fe3+) forms. The transformation reaction limits the deleterious reaction which occurs between ferrous iron and hydrogen peroxide, thereby inhibiting the production of damaging hydroxyl radical. In addition, ferritin is also involved in the regulation of infection or cancer. Ferritin levels are significantly increased in response to an infection or cancer. And endotoxins are an up-regulator of the gene coding for ferritin, thus inducing the concentration of ferritin to increase.

The Role of Ferritin in Sepsis
Sepsis is a form of systemic inflammatory response syndrome (SIRS) caused by an overwhelming reaction to infection, usually by bacteria. Ferritin is a positive acute phase reactant, however, in certain inflammatory diseases like Still's disease, it rises disproportionally high levels with respect to other acute-phase reactants, which suggest its possible contribution to the pathogenesis of disease during the active period of condition. Moreover, ferritin has been increased in children with septic shock and high ferritin levels are related to poorer outcome. Role of serum ferritin as a marker of disease activity and involvement severity has already been shown in sepsis, graft rejection, dermatomyositis, and SLE. Besides, serum ferritin is also a well-known inflammatory marker, but it is unclear whether serum ferritin reflects or causes inflammation, or whether it is involved in an inflammatory cycle. It has been considered that serum ferritin arises from damaged cells, and is thus a marker of cellular damage.
