Construction of 3D Ex Vivo Human Kidney Nephroblastoma Model

Since tumor cells have different growth characteristics and biological behaviors from normal kidney cells, establishing 3D culture models with characteristics of kidney nephroblastoma has become a powerful tool to study aspects of kidney disease progression and drug resistance mechanisms.

3D ex vivo Nephroblastoma Model

  • Case 1: Cell-matrix co-culture model.
    Nephroblastoma cells are inoculated in vitro in a stroma containing endothelial cells, fibroblasts, smooth muscle cells, and smooth muscle epithelial cells, which are passaged and differentiated to form the same 3D structure as nephroblastoma cells.
  • This model was mainly used to observe Nephroblastoma tissues and the vascular and lymphatic structures in them. By electron microscopic observation of this model, Nephroblastoma cells can be seen to be distinctly different from normal kidney tissues.

Multidirectional 3D Nephroblastoma Model

  • Case 2: matrix-scaffold-cell 3D co-culture model.
    In vitro inoculation of nephroblastoma cells on media containing different proportions of stroma or scaffold materials, after passaging and differentiation to form a 3D structure similar to that of kidney tumor tissue.
