Glioma is a common brain tumor characterized by its poor prognosis and strong aggressiveness. The rapid growth and frequent recurrence after surgical removal reflect its refractory nature. Normal cerebral organoids are a common type of normal neural organoids. With respect to their derivation, Xu et al. have verified a robust protocol. The normal cerebral organoids initiate from induced pluripotent stem cells (iPSC) and are next cultured into embryoid bodies. Through induction by a variety of growth factors, they gradually differentiate and finally generate cerebral tissues. The general culture process can be found in Fig.1.

Current research has found that it could make up for the shortcomings of traditional glioma models and displays the following advantages.
To our knowledge, normal cerebral organoids have been widely accepted as a good platform for glioma research since their emergence. They are very suitable for exploring glioma as well as its microenvironment, and can be employed to conduct disease modeling, toxicology testing, drug sensitivity test, personalized treatment and construct orthotropic xenografts. In view of the rapid development of microfluidic and organ-on-chip technology, such models display great potential for brain diseases in the near future.
