3D Ex Vivo Human Healthy GI Permeability Model Introduction

The human gastrointestinal tract (GI) is a dynamic, multilayered barrier that plays an important role in nutrient absorption and protection against the uptake of bacterial toxins. What's more, it is also responsible for the absorption and metabolism of drugs. Transport across the intestinal membrane is a complex process related to various functional pathways. Studies have shown that there are various factors governing drug absorption and intestinal permeability. Although a complex process, controlling oral drug absorption depends on drug permeability and solubility across the GI membrane. Ex Vivo Models for Healthy GI Permeability Research

During drug development, knowledge of GI absorption mechanisms can predict drug absorption and expected effects in the human intestine. In recent years, various models have been developed to characterize drug permeability. Compared with in vivo animal models, the in vitro approaches are always less labor-intensive and less expensive, and have ethical advantages. The tissue-based in vitro models, also known as ex vivo models, are obtained by isolating living functional tissues or organs from organisms and culturing them externally in a highly controlled environment. Nowadays, diffusion chambers are widely used for drug permeability study of intestinal tissue in vitro. In addition, Franz cell, everted intestinal sac, and everted intestinal ring are also suitable models for research.

Advantages of Ex Vivo Model for Permeability Study

  • Better simulation of in vivo conditions
  • Allows study of regional differences in intestinal absorption
  • Requires relatively small amounts of drug
  • Research of bidirectional drug transport
  • Tissue viability and integrity can be continuously monitored via electrical parameters
  • Allows for the research of drug permeability with different conditions (e.g., pH)
